pH6抗原是鼠疫菌毒力的重要组成部分,研究结果显示,这种抗原很可能与腺鼠疫的发病机制有关。为提供国内急需的pH6抗原纯品,研究和观察了该抗原的体外适宜表达条件。结果表明,在四种常规培基中,pH6抗原均可表达,但在LB培基中,该抗原的表达量明显高于其他培基。此外,还进行了两种pH及两种温度条件下该抗原表达量的比较,如预期的那样,该抗原仅在酸性和37℃条件下最大限度表达。根据该抗原不同时PH和温度条件下十二烷基磺酸钠—聚丙烯酰胺凝胶电泳(SDS—pAGE)图型的比较、参照已单体分子量,初步识别了该抗原的电泳图型,为今后进一步提取、纯化这种抗原成分奠定了基础。
The pH6 antigen is an important part of virulence of the plague bacteria and the results show that this antigen is likely to be related to the pathogenesis of bubonic plague. In order to provide the domestic demand of the pH6 antigen pure product, study and observe the appropriate expression conditions of the antigen in vitro. The results showed that the pH6 antigen was expressed in all four routine cultivars, but the expression level of the antigen in LB medium was significantly higher than that in other cultivars. In addition, a comparison of the expression level of the antigen under two pH and two temperature conditions was performed, and the antigen was maximally expressed only under acidic and 37C conditions as expected. According to the comparison of the sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) patterns at different PH and temperature conditions of the antigen, the electrophoretic pattern of the antigen was initially identified with reference to the molecular weight of the monomer, For the future further extraction, purification of this antigen composition laid the foundation.
