目的 :主要对高原土生动物 (鼠兔和鼢鼠 )与移居高原Wistar大白鼠肺灌洗液中肺表面活性物质成分及含量进行分析比较 ,从而探讨高原土生动物低氧适应机理及临床应用开发前景。方法 :所选用动物放血处死进行肺灌洗 ,得到的肺灌洗液采用FLONH方法萃取脂质并测定磷脂(TP)、磷脂酰甘油 (PG)、卵磷脂 (PC)和饱和卵磷脂 (SPC)。肺灌洗液经高速离心后按Bhattechar rya提取蛋白成分 ,粗提物采用紫外和克氏定氮法测定。结果 :Wistar大白鼠肺灌洗液中磷脂、磷脂酰甘油、卵磷脂和饱和卵磷脂低于高原鼠兔和鼢鼠 (P <0 .0 5 ) ;Wistar大白鼠蛋白质含量也低于这两种土生动物 (P <0 .0 5 ) ;高原鼠兔和鼢鼠含量基本接近。结论 :高原鼠兔和鼢鼠的肺表面活性物质在维持正常肺功能和适应高原缺氧起着重要的作用 ;Wistar大白鼠肺表面活性物质含量的降低标志着对高原低氧的不适应。
OBJECTIVE: To analyze and compare the composition and content of pulmonary surfactant in the lung lavage fluid of Wistar rats from plateau native animals (squirrel and zokor) and on the plateau, and to explore the mechanism of hypoxia adaptation and the prospect of clinical application and development of plateau indigenous animals . Methods: The animals were sacrificed by exsanguination and lung lavage was performed. The resulting lung lavage fluid was extracted by FLONH method and the contents of phospholipid (TP), phosphatidylglycerol (PG), lecithin (PC) and saturated lecithin (SPC) . Pulmonary lavage fluid by high-speed centrifugation Bhattechar rya extract protein components, crude extracts by UV and Kjeldahl determination. Results: Phospholipid, phosphatidylglycerol, lecithin and saturated lecithin in Wistar rat lung lavage fluid were lower than those in plateau pika and zokor (P <0.05); Wistar rats also had lower protein content The soil animals (P <0.05), the content of plateau pika and zokor were close to each other. CONCLUSIONS: Pulmonary surfactant in plateau pika and zokor plays an important role in maintaining normal lung function and adapting to plateau hypoxia. The decrease of pulmonary surfactant in Wistar rats indicates the incompatibility of plateau hypoxia.
