目的:观察开心解郁方对脑缺血大鼠学习记忆的影响,并探讨其作用机制。方法:Wistar大鼠分为假手术组、模型组和开心解郁方治疗组,采用双侧颈总动脉永久性阻断法复制3 d、7 d和21 d脑缺血模型,分别观察大鼠学习记忆能力、脑血流量变化和NO含量及NOS活性。结果:脑缺血3个时间点模型组大鼠,水迷宫游泳逃避潜伏期延长,额叶、顶叶的脑血流量显著减少,NO含量及NOS活性升高;而开心解郁方治疗组增加了额叶、顶叶脑血流量,缩短水迷宫游泳逃避潜伏期,NO含量及NOS活性在缺血3 d、7 d升高,21 d降低(P<0.05)。结论:开心解郁方对脑缺血大鼠学习记忆能力的改善可能与提高脑血流量、降低NO含量和NOS活性有关。
Objective: To observe the effect of Kaixin Jieyu Decoction on learning and memory in cerebral ischemia rats and to explore its mechanism. Methods: Wistar rats were divided into sham-operation group, model group and Kaixin Jieyu Decoction group. The bilateral common carotid artery occlusion method was used to replicate 3 d, 7 d and 21 d cerebral ischemia models. The learning and memory Ability, changes in cerebral blood flow and NO content and NOS activity. Results: In the cerebral ischemia group, swimming in the maze and swimming in the water maze prolonged the latent period of cerebral blood flow, the cerebral blood flow in the frontal lobe and parietal lobe decreased significantly, while the content of NO and the activity of NOS increased. In the Kai Xin Jie Yu decoction group, , Parietal lobe cerebral blood flow, shortened water maze swimming escape latency, NO content and NOS activity increased on the 3rd and 7th day of ischemia and decreased on the 21st day (P <0.05). Conclusion: The improvement of Kaixin Jieyu Decoction on learning and memory in rats with cerebral ischemia may be related to improving cerebral blood flow, decreasing NO content and NOS activity.
