分离的成年豚鼠心室肌单细胞在高钾(25mmol/L)溶液中部分除极化使钠通道失活,用细胞内刺激诱发慢反应动作电位。20μmol/L 的丹参酮Ⅱ-A 磺酸钠对这种慢反应动作电位有明显的抑制作用。在1—20μmol/L 浓度范围内,丹参酮Ⅱ-A 磺酸钠对0.28μmol/L 的异丙肾上腺素强化的慢反应动作电位有浓度依赖性抑制作用。而且,随异丙肾上腺素浓度的增加(69 nmol/L—0.55μmol/L)抑制作用更加明显。以上结果提示丹参酮 Ⅱ-A 磺酸钠可能是一种钙拮抗剂。50—100μmol/L 的丹参酮 Ⅱ-A 磺酸钠使分离的成年豚鼠心室肌单细胞快反应动作电位的幅度降低,达峰值的时间延长。提示高浓度的丹参酮 Ⅱ-A 磺酸钠对钠通道也有一定的阻断作用。
Isolated adult guinea pig ventricular myocytes in high potassium (25mmol / L) solution partial depolarization of the sodium channel inactivation of intracellular stimuli induced slow response action potential. 20μmol / L sodium tanshinone Ⅱ-A sulfonate slow response to this action potential was significantly inhibited. In the concentration range of 1-20 μmol / L, tanshinone Ⅱ-A sulfonate inhibited concentration-dependently the action potential of 0.28 μmol / L isoproterenol-potentiated slow reaction. Moreover, the inhibitory effect with isoproterenol concentration (69 nmol / L-0.55 μmol / L) was more pronounced. The above results suggest that tanshinone Ⅱ-A sulfonate may be a calcium antagonist. 50-100μmol / L sodium tanshinone Ⅱ-A sodium salt isolated adult guinea pig ventricular myocytes rapid response action potential amplitude decreased, reaching the peak time extension. Tip high concentrations of sodium tanshinone Ⅱ-A sodium channel also has some blocking effect.
