目的:基于众多文献报道MTT法化疗药敏测定中癌细胞抑制率与外周血淋巴细胞抑制率呈正相关,探讨预制检测板MTT法在检测癌患者外周血淋巴细胞药物敏感性中的价值,以推动检测标准化与指导临床化疗用药。方法:应用泰伦公司生产的MTT法肿瘤细胞药敏预制检测板,对75例癌患者外周血淋巴细胞进行20种药物的药敏检测,操作方法按预制检测板使用说明书。结果:39例肺癌患者外周血淋巴细胞对化疗药物敏感性,由高到低前5名依次为:卡氮芥(BCNU),紫杉醇(PTX),阿霉素(ADM),长春瑞滨(NVB)和表阿霉素(eADM);25例乳腺癌患者外周血淋巴细胞对化疗药物敏感性,前5名依次为:BCNU,PTX,NVB,ADM和米托蒽醌(MIT);11例胃癌患者外周血淋巴细胞对化疗药物敏感性,前5名依次为:PTX,ADM,BCNU,eADM和阿糖胞苷(Ara-C)。结论:该MTT法预制检测板适用于癌患者药物敏感性试验,对临床癌患者个体化化疗有重要指导意义。
OBJECTIVE: To evaluate the value of MTT assay in the detection of drug sensitivity of peripheral blood lymphocytes in patients with cancer by the MTT assay, which is based on the positive correlation between the inhibitory rate of cancer cells and the rate of peripheral blood lymphocyte inhibition in MTT assay. Methods: The antimicrobial susceptibility testing kit of MTT method was used to detect the drug susceptibility of 20 kinds of drugs in peripheral blood lymphocytes of 75 cases of cancer patients. The operation method was according to the manual of prefabricated test plate. Results: The sensitivity of chemotherapeutics to peripheral blood lymphocytes in 39 patients with lung cancer was BCNU, PTX, ADM, vinorelbine (NVB) ) And epirubicin (eADM). The chemosensitivity of peripheral blood lymphocytes in 25 patients with breast cancer were top 5: BCNU, PTX, NVB, ADM and mitoxantrone (MIT); 11 cases of gastric cancer The sensitivity of chemotherapeutic drugs to peripheral blood lymphocytes in patients was as followed: PTX, ADM, BCNU, eADM and Ara-C. Conclusion: The MTT prefabricated test plate is suitable for the drug sensitivity test of cancer patients, which has important guiding significance for individualized chemotherapy in patients with clinical cancer.
