目的:探讨己烯雌酚对去卵巢大鼠股骨颈松质骨的影响。方法:采用3月龄SPF级SD雌性大白鼠33只,随机分为4组:基础对照组、假手术组、去卵巢组和去卵巢+DES治疗组,后者灌胃4.5mg.L-1的己烯雌酚(5ml.kg-1.d-1),每周6次,90d后处死,取股骨颈经不脱钙骨制片进行骨组织形态计量学参数测量。结果:与对照组比较,去卵巢大鼠静态参数的骨小梁面积百分数和骨小梁数量分别减少了35.5%(P<0.01)和27.3%(P<0.01),骨小梁间隙增大了83.9%(P<0.01);动态参数的每毫米破骨细胞数和破骨细胞贴壁表面长度分别增加了130.0%(P<0.01)和181.0%(P<0.01),骨矿化沉积率则减少了26.3%(P<0.01)。与去卵巢组比较,去卵巢+DES组动态参数每毫米破骨细胞数和破骨细胞贴壁表面长度分别减少了43.4%(P<0.05)和47.6%(P<0.05),骨矿化沉积率、单位骨小梁周长骨形成率和单位骨小梁面积骨形成率也分别减少了27.2%(P<0.01)、58.0%(P<0.05)和58.1%(P<0.05);静态参数骨小梁面积、数量分别增加了19.0%和13.3%(尽管其差异无显著性)。结论:大鼠去卵巢后能导致大鼠股骨颈骨量显著减少,己烯雌酚则能阻止去卵巢所致大鼠股骨颈骨量丢失。
Objective: To investigate the effect of diethylstilbestrol on cancellous bone of femoral neck in ovariectomized rats. Methods: Thirty-three SD female SD rats of 3 months old were randomly divided into 4 groups: basal control group, sham operation group, ovariectomized group and ovariectomized + DES treated group. Of diethylstilbestrol (5ml.kg-1.d-1), 6 times a week, 90d after sacrifice, take the femoral neck of non-decalcified bone preparation bone histomorphometry parameters measurement. Results: Compared with the control group, the static trabecular area percentage and trabecular number of ovariectomized rats decreased by 35.5% (P <0.01) and 27.3% (P <0.01), respectively, and the trabecular clearance increased 83.9%, respectively (P <0.01). The number of osteoclasts per mm in dynamic parameters and the length of adherent osteoclasts increased by 130.0% (P <0.01) and 181.0% Decreased by 26.3% (P <0.01). Compared with the ovariectomized group, the dynamic parameters of ovariectomized + DES group decreased by 43.4% (P <0.05) and 47.6% (P <0.05) respectively, while the number of osteoclasts per mm and osteoclast adherent surface length decreased by The rates of bone formation per unit trabecular bone and bone formation per trabecular area were also decreased by 27.2% (P <0.01), 58.0% (P <0.05) and 58.1% (P <0.05) Trabecular area and number increased by 19.0% and 13.3% respectively (although the difference was not significant). CONCLUSION: Ovariectomized rats can significantly reduce the femoral neck mass and diethylstilbestrol can prevent femoral neck bone loss in ovariectomized rats.
