目的探讨RhoA特异性抑制剂C3胞外酶对乙醇诱导的肠上皮细胞通透性增加过程中的抑制作用。方法予乙醇及C3胞外酶作用肠上皮细胞屏障后,测定跨上皮细胞电阻值(TEER),分析肠上皮细胞屏障的通透性的变化。应用Western blot检测紧密连接跨膜蛋白occludin的表达;应用免疫荧光染色法检测紧密连接蛋白occludin表达和分布。结果予C3胞外酶预处理后,可显著抑制乙醇诱导的TEER值降低及occludin(S/IS)比值的增加,同时可部分恢复细胞膜上occludin的表达。结论 RhoA特异性抑制剂C3胞外酶可抑制乙醇诱导肠上皮细胞屏障通透性增加的过程。
Objective To investigate the inhibitory effect of RhoA-specific inhibitor C3 extracellular enzyme on ethanol-induced increase of intestinal epithelial cell permeability. Methods After the barrier of intestinal epithelial cells was treated with ethanol and C3 extracellular enzyme, the transmembrane cell resistance (TEER) was measured and the permeability of intestinal epithelial barrier was analyzed. Western blot was used to detect the expression of occludin. The expression of occludin and its distribution were detected by immunofluorescence staining. Results Pretreatment with C3 extracellular enzyme could significantly inhibit the decrease of ethanol induced TEER and the increase of occludin (S / IS) ratio, and partially restore the expression of occludin on the cell membrane. Conclusion RhoA-specific inhibitor C3 extracellular enzyme can inhibit ethanol-induced intestinal epithelial cell barrier permeability increased process.
