参照杨连甲的方法从小牛骨基质中提取牛骨形成蛋白(bBMP),经SDS-PAGE电泳分析,bBMP含67、43、30、22、18和14kD六种蛋白组分,保留BMP活性蛋白成份,植入小鼠股部肌肉后的X线及组织学观察发现,所提取的bBMP具有较高的骨诱导活性,bBMP植入后14天即可见有新骨形成,21天时有含骨髓的板层状骨形成。采用bBMP单克隆抗体的免疫组织化学染色发现,BMP存在于未分化间充质细胞胞浆和骨形成过程中的成骨细胞内。
BbMP was extracted from the bovine bone matrix by reference to the method of Yang Lian Jia. BBMP contained 67, 43, 30, 22, 18 and 14 kD protein components by SDS-PAGE electrophoresis and retained the protein components of BMP activity, X-ray and histological observation showed that the extracted bBMP had higher osteoinductive activity, and new bone formation was seen at 14 days after bBMP implantation. On the 21st day, there was bone marrow-lamellar layer Bone formation. Immunohistochemical staining with bBMP monoclonal antibody showed that BMP was present in osteoblasts in the cytoplasm and bone formation of undifferentiated mesenchymal cells.
