目的:探讨滋补脾阴方药(ZiBu PiYin Recipe,ZBPYR)对脾阴虚糖尿病大鼠海马内质网应激相关分子表达的影响。方法:高脂饲料喂养联合小剂量链脲佐菌素(streptozocin,STZ)注射建立2型糖尿病模型,在此基础上,通过复合因素造模的方法建立脾阴虚糖尿病模型,以水迷宫实验评价模型大鼠是否出现学习记忆障碍,之后观察各组大鼠海马内质网应激相关分子葡萄糖调节蛋白78(glucose regulated protein 78,GRP78)、蛋白激酶样内质网激酶(protein kinase RNA-like endoplasmic reticulum kinase,PERK)和真核转录因子2的α亚单位(eukaryotic translation initiation factor 2 subunitα,eIF2α)表达的变化。结果:ZBPYR治疗后GRP78 mRNA水平降低(P<0.05),GRP78、磷酸化PERK、磷酸化eif2α蛋白水平明显降低(P<0.05),与脾阴虚糖尿病组相比,差异有统计学意义。结论:滋补脾阴方药可能是通过影响内质网应激PERK信号传导改善脾阴虚糖尿病大鼠学习记忆障碍。
Objective: To investigate the effects of ZiBu PiYin Recipe (ZBPYR) on the endoplasmic reticulum stress-related molecules in hippocampus of spleen yin-deficiency rats. Methods: Type 2 diabetes mellitus was induced by high-fat diet combined with low-dose streptozocin (STZ) injection. On the basis of this, diabetes model of spleen-yin deficiency was established by composite factor modeling and evaluated by water maze test To investigate whether learning and memory impairment occurs in the model rats, and then observe the changes of hippocampal ER stress-related molecules, glucose regulated protein 78 (GRP78), protein kinase RNA-like endoplasmic reticulum kinase (PERK) and eukaryotic translation initiation factor 2 subunitα (eIF2α). Results: After treatment with ZBPYR, GRP78 mRNA level decreased (P <0.05), GRP78, phosphorylated PERK and phosphorylated eif2α protein levels decreased significantly (P <0.05). Compared with spleen yin deficiency diabetes group, the difference was statistically significant. Conclusion: Nourishing spleen Yin prescription may improve learning and memory impairment of rats with spleen-yin deficiency syndrome by affecting PERK signaling in endoplasmic reticulum stress.
