目的:优化超临界CO2萃取穗花大黄中总蒽醌的工艺条件。方法:以萃取压力、萃取温度和萃取时间为考察因素,采用紫外-可见分光光度法测定总蒽醌含量,以大黄萃取物中总蒽醌的含量为评价指标,正交试验优选工艺条件。结果:最佳工艺条件为萃取压力25 MPa,萃取温度50℃,萃取2.0 h。提取物中总蒽醌的质量分数23.4 mg.g-1,总蒽醌转移率达91.4%。结论:该工艺稳定可行,可用于穗花大黄总蒽醌的提取。
Objective: To optimize the technological conditions of total anthraquinone extraction from Panax rhizome by supercritical CO2. Methods: The extraction pressure, extraction temperature and extraction time were investigated. The content of total anthraquinone was determined by UV-Vis spectrophotometry. The content of total anthraquinone in rhubarb extract was used as evaluation index. The orthogonal experiment was used to optimize the process conditions. Results: The optimum conditions were extraction pressure 25 MPa, extraction temperature 50 ℃ and extraction 2.0 h. The mass fraction of total anthraquinone in the extract was 23.4 mg.g-1, and the total anthraquinone transfer rate was 91.4%. Conclusion: The process is stable and feasible, which can be used for the extraction of total anthraquinones from Rheum.
