目的:观察健延龄胶囊对正常雄性大鼠以及环磷酰胺所致少、弱精雄性大鼠精子活力及质量的影响。方法:选取40只SD大鼠随机分为正常对照组、健延龄(0.8、1.6、3.2g/kg)给药组,除正常对照组灌胃给予生理盐水外,其余各组均灌胃给予健延龄,连续30天。给药结束后次日上午脱颈椎处死大鼠,取右侧完整附睾进行精子活力检测,并通过HE染色观察各组大鼠左侧睾丸与附睾病理形态学改变。另外,通过腹腔注射环磷酰胺方法制备少、弱精雄性大鼠模型,选取50只SD大鼠随机分为:正常对照组、模型对照组、健延龄(0.8、1.6、3.2g/kg)给药组,除正常对照组外,其余各组动物每只腹腔注射环磷酰胺0.3mg/kg/d进行造模,连续5天,第6天开始各给药组开始灌胃给予健延龄,正常对照组和模型对照组灌胃给予生理盐水,连续30天。于给药结束后次日上午脱颈椎处死大鼠,取右侧完整附睾进行精子活力检测,并通过HE染色观察各组大鼠左侧睾丸与附睾病理形态学改变。结果:健延龄1.6 g/kg对正常大鼠的精子运动速度有促进作用,显著增加正常大鼠的精子数量,提高正常大鼠的精子浓度;组织学检测显示,健延龄0.8 g/kg、1.6 g/kg和3.2 g/kg对正常大鼠睾丸组织及附睾组织均无明显影响;环磷酰胺注射后的大鼠经健延龄1.6 g/kg、3.2 g/kg给药后,能够显著增强精子的运动速度,明显增加精子数量;组织学检测显示,环磷酰胺所致大鼠少、弱模型,在给予健延龄0.8 g/kg、1.6 g/kg和3.2 g/kg药物后,发现个别大鼠睾丸曲细精管内生精细胞排列轻度紊乱,附睾管内精子数量轻微减少,且组织学评分结果为:模型组0.7分,健延龄0.8 g/kg组0.28分,健延龄1.6 g/kg组0.25分,健延龄3.2 g/kg组0.11分。结论:健延龄胶囊对正常大鼠的精子运动、精子密度方面有一定的增强作用;健延龄胶囊给药后,对环磷酰胺引起的大鼠精子运动减弱及数量减少等现象有明显的改善作用;健延龄对环磷酰胺所致大鼠少、弱精症睾丸的病理改变有明显改善作用。
