采用肾上腺素法测定了超氧化物歧化酶的酶活,确定了最佳测定波长,并研究各种实验条件对超氧化物歧化酶活性的影响。结果表明最优的测定酶活条件为:测定波长为490 nm,实验温度为30℃,0.05 mol·L-1碳酸钠-碳酸氢钠内含1×10-4mol·L-1乙二胺四乙酸缓冲溶液(pH为10.2),肾上腺素浓度为3×10-4mol·L-1,控制酶的加入量为0.30 mL。
The enzyme activity of superoxide dismutase (SOD) was determined by adrenaline method, the best determination wavelength was determined, and the effect of various experimental conditions on superoxide dismutase activity was studied. The results showed that the optimum conditions for the determination of enzyme activity were as follows: the measurement wavelength was 490 nm, the experimental temperature was 30 ℃, 0.05 mol·L -1 sodium carbonate-sodium bicarbonate contained 1 × 10-4 mol·L-1 ethylenediamine tetra Acetate buffer solution (pH 10.2), epinephrine concentration 3 × 10-4mol·L-1, control enzyme was added in an amount of 0.30mL.
