目的:探讨组蛋白去乙酰化酶1(HDAC1)蛋白在早期自然流产发生中的病理生理作用及临床意义。方法:应用免疫组织化学和Western blot法检测早期自然流产和早期正常妊娠者绒毛及蜕膜组织中HDAC1蛋白的表达。结果:免疫组化示:HDAC1主要定位于绒毛细胞滋养细胞、合体滋养细胞和子宫蜕膜细胞、腺上皮细胞的胞核;早期自然流产绒毛组织中HDAC1的表达强度低于早期正常妊娠组,而蜕膜组织中,早期自然流产组织HDAC1的表达强度高于正常妊娠组,差异均有统计学意义(P均<0.05)。Westernblot示:早期自然流产绒毛组织中HDAC1蛋白的相对表达量为1.38±0.63,明显低于正常妊娠组(1.92±0.82),差异有统计学意义(P<0.05);而早期自然流产蜕膜组织中HDAC1蛋白的相对表达量为1.02±0.53,明显高于正常妊娠组(0.71±0.36),差异有统计学意义(P<0.05)。结论:HDAC1在早期自然流产绒毛和蜕膜组织中的异常表达可能在早期自然流产的发生、发展中起重要作用。
Objective: To investigate the pathophysiological role of histone deacetylase 1 (HDAC1) protein in early spontaneous abortion and its clinical significance. Methods: The expressions of HDAC1 protein in villus and deciduas of early spontaneous abortion and early normal pregnancy were detected by immunohistochemistry and Western blot. Results: Immunohistochemistry revealed that HDAC1 mainly located in the nucleus of villous trophoblasts, syncytiotrophoblasts, uterine decidual cells and glandular epithelial cells. The expression intensity of HDAC1 in early spontaneous abortion villus was lower than that in early normal pregnancy group In decidua, the expression intensity of HDAC1 in early spontaneous abortion group was higher than that in normal pregnancy group (all P <0.05). Westernblot showed that the relative expression level of HDAC1 protein in early spontaneous abortion chorion was 1.38 ± 0.63, which was significantly lower than that in normal pregnancy group (1.92 ± 0.82) (P <0.05), whereas the early spontaneous abortion decidual tissue The relative expression level of HDAC1 protein was 1.02 ± 0.53, which was significantly higher than that of normal pregnancy group (0.71 ± 0.36), the difference was statistically significant (P <0.05). Conclusion: The abnormal expression of HDAC1 in early spontaneous abortion villus and decidua may play an important role in the occurrence and development of early spontaneous abortion.
