利用抗体-抗原-酶标抗体反应,建立了间接酶联免疫吸附测定法(ELISA),以检测转基因抗虫棉品系310幼叶Bt毒蛋白含量。初步测定结果:转基因抗虫棉幼叶中Bt毒蛋白含量为总可溶性蛋白质的(0.126±0.008)%。提纯的伴胞晶体样品经ELISA法检测,其Bt毒蛋白最低可检值为20ng/ml。这一检测技术既可对转基因抗虫棉组织中Bt毒蛋白表达量进行定性、定量测定,也为阐明转基因抗虫棉的抗虫特性提供了理论依据。
Indirect enzyme-linked immunosorbent assay (ELISA) was developed to detect Bt toxin content in young leaves of transgenic Bt cotton line 310 using antibody-antigen-enzyme antibody reaction. Preliminary results showed that Bt protein content in transgenic Bt cotton was (0.126 ± 0.008)% of total soluble protein. The purified companion crystal samples were detected by ELISA. The lowest detectable value of Bt toxin was 20ng / ml. The detection technology can not only qualitative and quantitative determination of Bt toxin protein expression in transgenic Bt cotton tissue, but also provide a theoretical basis for elucidating the insect-resistant properties of transgenic Bt cotton.
