目的研究急性长时间运动时血浆游离脂肪酸(FFA)对骨骼肌线粒体解偶联蛋白3(UCP3)表达的影响。方法雄性SD大鼠30只随机分成安静对照组(RC),脂解抑制剂(烟酸)运动组(NE)和安慰剂运动组(PE)。后两组进行时间为150 min的递增负荷跑台运动,观察运动前后血浆FFA、UCP3 mRNA及骨骼肌和线粒体UCP3蛋白表达的变化。结果 PE组和NE组血浆FFA、UCP3 mRNA及骨骼肌和线粒体UCP3蛋白均显著高于RC组;NE组血浆FFA、UCP3mRNA低于PE组(FFA:0.49±0.05 vs 0.67±0.07 mmol/L,(F=153.850),P﹤0.05;UCP3 mRNA:5.49±0.71 vs9.60±1.15,(F=79.877),P﹤0.01),但骨骼肌和线粒体UCP3蛋白与PE组差异无统计学意义(骨骼肌UCP3:1.31±0.12 vs 1.38±0.16,(F=20.844),P﹥0.05;线粒体UCP3:1.45±0.17 vs 1.42±0.15,(F=47.882),P﹥0.05)。结论 FFA可能参与了急性长时间运动时骨骼肌线粒体UCP3的表达上调。
Objective To investigate the effect of plasma free fatty acid (FFA) on the expression of uncoupling protein 3 (UCP3) in skeletal muscle during acute prolonged exercise. Methods Thirty male Sprague - Dawley rats were randomly divided into three groups: control group (RC), lipolytic (niacin) exercise group (NE) and placebo group (PE). The latter two groups were performed treadmill exercise with increasing load for 150 min. The changes of plasma FFA, UCP3 mRNA and skeletal muscle and mitochondrial UCP3 protein expression were observed before and after exercise. Results The levels of plasma FFA, UCP3 mRNA and skeletal muscle and mitochondrial UCP3 protein in PE group and NE group were significantly higher than those in RC group. The levels of FFA and UCP3 mRNA in NE group were lower than those in PE group (FFA: 0.49 ± 0.05 vs 0.67 ± 0.07 mmol / L, (F = 153.850), P <0.05; UCP3 mRNA: 5.49 ± 0.71 vs 9.60 ± 1.15, (F = 79.877, P <0.01). However, there was no significant difference between skeletal muscle and mitochondrial UCP3 protein in PE group UCP3: 1.31 ± 0.12 vs 1.38 ± 0.16, (F = 20.844), P> 0.05; mitochondrial UCP3: 1.45 ± 0.17 vs 1.42 ± 0.15, (F = 47.882, P> 0.05). Conclusion FFA may be involved in the up-regulation of skeletal muscle mitochondrial UCP3 during acute long-term exercise.
