目的研究X线诱导两种人鼻咽癌细胞系凋亡的情况。材料与方法应用DNA特异荧光染料Hoechst33342观察凋亡细胞,细胞的照射应用直线加速器。结果给一固定剂量照射后,CNE细胞系凋亡指数与时间和剂量成相关性,于照射后48小时达到平台期。在分次放疗中,CNE细胞系总剂量10Gy,照射5次/5天组的凋亡指数较照射2次/5天和照射1次/1天组高,分别为37.8%、34.5%和22.8%,2Gy照射后于24小时凋亡指数恢复至初始水平,而5Gy至少需要72小时。分别照射0,2,4,8,12和20Gy后CNE凋亡指数分别为2.0%、10.2%、17.8%、21.4%、24.0%、27.0%;CNE2的分别为3.5%、15.0%、20.0%、31.0%、38.0%、48.0%。结论人鼻咽癌细胞系的凋亡指数与时间和剂量呈相关性,照射后凋亡水平恢复的时间与照射剂量密切相关。两种细胞系的凋亡指数存在明显差异。
Objective To investigate the apoptosis of human nasopharyngeal carcinoma cells induced by X-rays. Materials and Methods Apoptotic cells were observed using DNA-specific fluorescent dye Hoechst33342. The cells were irradiated with a linear accelerator. Results After a fixed dose of irradiation, the apoptotic index of CNE cell line was correlated with time and dose, reaching plateau 48 hours after irradiation. In fractionated radiotherapy, the apoptotic index of the CNE cell line at a dose of 10 Gy and 5/5 days was 37.8% and 34.8%, respectively, compared with that of 2 and 5 days and 1 and 1 day respectively. 5% and 22.8%. After 2 Gy irradiation, the apoptotic index recovered to the initial level at 24 hours, while it took at least 72 hours for 5 Gy. The apoptotic indexes of CNE were 2.0%, 10.2%, 17.8%, 21.4%, 24.0% and 27.0% at 0, 2, 4, 8, 12 and 20 Gy respectively. CNE 2 were 3.5%, 15.0%, 20.0%, 31.0%, 38.0%, 48.0% respectively. Conclusion The apoptotic index of human nasopharyngeal carcinoma cell line is correlated with time and dose. The time of recovery of apoptosis level after irradiation is closely related to the irradiation dose. There was a significant difference in apoptosis index between the two cell lines.
