用结晶紫染色法、3~H-TdR掺入、增殖细胞核抗原(PCNA)免疫细胞化学法检测3,4-二羟基苯乙酮(3,4-DHAP)对缺氧所致猪肺动脉平滑肌细胞(PASMC)增殖的影响。结果发现缺氧条件下培养的PASMC用以上3种方法测得结果分别为常氧组的(117.1±4.7)%、(131.2±17.0)%、(112.8±6.2)%,均有显著性差异(均为P<0.01);3,4-DHAP对缺氧所致PASMC增殖的3项指标变化均有明显抑制作用,并呈剂量依赖关系。因PASMC增殖是引起肺血管构型改建、导致慢性肺动肺高压的主要发病因素,故3,4-DHAP有可能用于慢性肺动脉高压的防治。
The effects of 3,4-dihydroxyacetophenone (3,4-DHAP) on hypoxia-induced pulmonary arterial smooth muscle cells (HUVECs) were detected by crystal violet staining, 3 H-TdR incorporation and proliferating cell nuclear antigen (PCNA) (PASMC) proliferation. The results showed that the PASMC cultured under hypoxia conditions were significantly (117.1 ± 4.7)%, (131.2 ± 17.0)% and (112.8 ± 6.2)%, respectively, compared with the normoxia group All P <0.01). 3,4-DHAP significantly inhibited the proliferation of PASMC induced by hypoxia in a dose-dependent manner. 3,4-DHAP may be used for the prevention and treatment of chronic pulmonary hypertension because proliferation of PASMC is a major causative factor for the alteration of the pulmonary vascular architecture that leads to chronic pulmonary hypertension.
