根据反转录转座子的反转录酶(reversetranscriptase,RT)区域序列特征,研究了小麦基因组中反转录转座子家族构成及其进化关系.根据反转录转座子RT保守序列设计引物,通过RTPCR从接种白粉病菌的小麦品系Pm97034的cDNA中扩增了一批RT片段.对51个随机克隆(RT1~51)的测序结果表明:它们之间具有高度异质性,核苷酸的同源性在75.4%~97.9%.结合已报道的7个小麦反转录转座子家族(family1~7)的代表序列,构建了系统发育树,发现3个新的家族,分别命名为family8,family9和family10.其中,family8是发生Pooideae亚族分化之前,是一类古老的反转录转座子家族.另外,family4和family7这2个家族中某些成员具有转录活性,并以高拷贝的形式存在于小麦及其近缘属的基因组中.
Based on the sequence characteristics of retrotransposase (RT) region of retrotransposon, the family structure of retrotransposon in wheat genome and its evolutionary relationship were studied.According to the conserved sequence of retrotransposon RT A series of RT fragments were amplified from the cDNA of Pm97034 wheat strain inoculated with powdery mildew by RTPCR.The sequencing results of 51 random clones (RT1 ~ 51) showed that they were highly heterogeneous and the nucleotide (75.4% -97.9%). Based on the reported sequences of 7 families of wheat transposon families (family1-7), the phylogenetic tree was constructed and three new families were found, named as family8, family9 and family10, of which family8 is an ancient family of retrotransposons prior to the Pooideae subfamily differentiation.In addition, some members of family 4 and family7, which are transcriptionally active, The form of a copy exists in the genome of wheat and its close relatives.
