研究建立了一套适于我国北方马铃薯主栽品种东农303外源基因导入的体系,并成功地将烟草病原蛋白基因导入该品种.结果显示,块茎外植体比其它外植体更适用于基因导入。农杆菌浓度对T—DNA转移有明显影响,5×108CFU/mL是较适宜的浓度.组织化学和分子印迹杂交证明,外源基因已经整合入马铃薯染色体组中。试验选出8个转基因个体,将通过无性系繁殖,进一步鉴定遗传稳定性。
We have established a system suitable for the introduction of foreign gene of Dongnong 303, a potato major cultivar in northern China, and successfully introduced the tobacco pathogen protein gene into this variety. The results showed that tuber explants were more suitable for gene introduction than other explants. Agrobacterium tumefaciens concentration has a significant impact on T-DNA transfer, 5 × 108CFU / mL is the more appropriate concentration. Histochemistry and molecular blot hybridization proved that exogenous genes have been integrated into the potato genome. Eight transgenic individuals were selected and were tested for their genetic stability by clonal propagation.
