目的:探索和络泄浊颗粒对大鼠肾脏纤维化中miR-200a表达的影响及探讨其可能存在的作用机制。方法:30只SD雄性大鼠随机分为6组:假手术组(Sham)、手术组(UUO)及和络泄浊颗粒(UUO+REG)组各2组,运用单侧(左)输尿管结扎法制造肾间质纤维化模型,但Sham组仅游离输尿管,而其余两组则游离并结扎输尿管。术后各组按1 mg/kg·d-1的量进行灌胃,UUO+REG组给予REG,其余两组则予生理盐水。术后第7天和14天,摘除左梗阻侧肾脏进行免疫组化检查α-SMA和荧光定量PCR检测miR-200a的表达。结果:在UUO及UUO+REG组,α-SMA表达明显高于Sham组(P<0.01);UUO+REG组低于UUO组(P<0.05)。miR-200a表达水平在UUO组和UUO+REG组都是降低的,但是UUO组与Sham组差别明显(P<0.01),其与UUO+REG组亦有明显的差别(P<0.05)。结论:和络泄浊颗粒可以通过上调miR-200a延缓肾脏纤维化进展。
Objective: To explore the effect of Hesuxiaoyu particles on the expression of miR-200a in rat kidney fibrosis and to explore its possible mechanism. Methods: Thirty male Sprague-Dawley rats were randomly divided into 6 groups: sham group, UUO group and UUO + REG group. Each group was divided into two groups by unilateral (left) ureteral ligation The model of renal interstitial fibrosis was made, but the Sham group only free ureter, while the remaining two groups were free and ligation of the ureter. The rats in each group were given gavage at the dose of 1 mg / kg · d-1, while those in the UUO + REG group were given REG. The other two groups were given saline. On the 7th day and the 14th day after operation, the left obstruction side of the kidney was removed and the expression of miR-200a was detected by α-SMA and fluorescence quantitative PCR. Results: The expression of α-SMA in UUO and UUO + REG groups was significantly higher than that in Sham group (P <0.01). The UUO + REG group was lower than UUO group (P <0.05). The expression of miR-200a was decreased in UUO group and UUO + REG group, but the difference between UUO group and Sham group was significant (P <0.01). There was also significant difference between UUO group and UUO + REG group (P <0.05). Conclusion: Huxuxie particles can delay the progression of renal fibrosis by up-regulating miR-200a.
