目的:探讨苍术麸炒前后氯仿部位的化学成分变化,从化学成分和药效学的变化角度探讨苍术的炮制机制。方法:运用HPLC技术研究苍术麸炒前后氯仿部位整体的化学成分,分离炮制前后变化明显的目标化合物;以健脾和燥湿为指标研究该部位炮制前后药效变化。结果:从氯仿部位分离得到2个变化明显的化学成分,化合物A为5-羟甲基糠醛,化合物B为邻苯二甲酸二异丁酯;药理实验结果表明,麸炒苍术氯仿部位组胃泌素水平高于生品,小肠推进率显著增加。各给药组尿量与尿液中AQP2含量变化较阴性对照组变化不明显。结论:变化成分邻苯二甲酸二异丁酯和5-羟甲基糠醛可能为麸炒苍术毒副作用和药效变化的物质基础之一;氯仿部位是健脾有效部位之一,该部位燥湿作用不明显。
OBJECTIVE: To investigate the chemical composition changes of chloroform fraction before and after herb bran extraction, and discuss the processing mechanism of herb herb from the perspective of chemical composition and pharmacodynamics. Methods: The HPLC method was used to study the chemical constituents of the whole chloroform fraction before and after herb bran extraction. The target compounds with obvious changes before and after processing were isolated. The effects of spleen and dampness on the pharmacodynamics of the parts were studied. Results: Two chemical constituents with obvious changes were obtained from the chloroform fraction. Compound A was 5-hydroxymethylfurfural and compound B was diisobutyl phthalate. Pharmacological results showed that the contents of gastrin The level of hormone is higher than that of raw product, and the rate of intestinal propulsion is increased significantly. The amount of urine in each administration group and urine AQP2 content changes than the negative control group did not change significantly. CONCLUSION: Diisobutyl phthalate and 5-hydroxymethylfurfural may be one of the material bases for the side effects and pharmacodynamic changes of Cortex Atractylodes; Chloroform is one of the effective parts of spleen, The role is not obvious.
