两株不同转移力小鼠肝癌细胞对宿主淋巴系统的影响

目的 研究高低转移力小鼠肝癌细胞 HCa- F和 HCa- P对宿主淋巴系统的影响及其机制。方法 以流式细胞仪动态检测皮下接种 F或 P细胞后的 6 15小鼠局部引流淋巴结内淋巴细胞的细胞周期。以免疫组化法检测F和 P细胞表达 Fas L 的情况 ,以 TU NEL 法观察转移淋巴结内巨噬细胞的凋亡情况。结果  F组小鼠引流淋巴结内淋巴细胞增殖峰出现晚于 P组 ,且持续时间短 ,迅速滑落至低位。F细胞表达 Fas L 强于 P细胞。F细胞周围的巨噬细胞发生凋亡。结论 宿主对 F细胞的免疫应答弱于 P。这可能与 F细胞表达较多 Fas L 诱导巨噬细胞凋亡 ,从而抑制抗原提呈作用有关。 Objective To investigate the effect of high and low metastatic mice liver cancer cells HCa-F and HCa-P on the host lymphatic system and its mechanism. Methods Flow cytometry was used to detect the cell cycle of lymphocytes in local draining lymph nodes of 6 15 mice subcutaneously vaccinated with F or P cells. The expression of Fas L in F and P cells was detected by immunohistochemistry and the apoptosis of macrophages in metastatic lymph nodes was observed by TU NEL. Results The lymphocyte proliferation peak in draining lymph nodes of group F appeared later than that of group P, and the duration was short and quickly slipped to a low level. F cells express Fas L stronger than P cells. Apoptosis occurs in macrophages surrounding F cells. Conclusion The immune response of the host to F cells is weaker than P. This may be related to the fact that F cells express more Fas L to induce macrophage apoptosis and thus inhibit antigen presentation.
