免疫亲和层析法纯化苦瓜几丁酶

用扁豆几丁酶免疫家兔,获得抗扁豆几丁酶的抗体,将此抗体与Sepharose 4B偶联,制备免疫亲和吸附剂,用以纯化苦瓜几丁酶.苦瓜叶片的粗提液经过免疫亲和吸附柱后,可获得电泳纯的几丁酶,其分子量为35 kD,与用几丁质凝胶为亲和吸附剂的纯化结果一致.表明利用植物几丁酶在结构上的保守性,用免疫亲和法可纯化不同植物的同类几丁酶.与几丁质凝胶亲和柱相比,免疫亲和法纯化植物几丁酶具有快速、亲和柱可重复使用等的优点.利用免疫亲和层析获得的纯化样品,研究了苦瓜几丁酶对真菌的抑制试验,研究结果表明,苦瓜几丁酶能分解棉花枯萎病菌的菌丝体细胞壁制备物,并对其孢子芽管的伸长有一定抑制作用. The lentigrin was used to immunize rabbits to obtain the anti-lentil enzyme antibody. The antibody was coupled with Sepharose 4B to prepare immunoaffinity sorbent for purification of bitter gourd chitinase. The crude extract of bitter gourd leaf after immunoaffinity adsorption column, electrophoresis can be pure chitinase enzyme, the molecular weight of 35 kD, with the chitosan gel affinity adsorbent purification results. Show that the use of plant chitinase in the structural conservation, immunoaffinity purification of different plants can be similar chitinase. Compared with chitosan gel affinity column, immunoaffinity purification of plant chitinase has the advantages of fast, affinity column reusability and so on. The purified samples obtained by immunoaffinity chromatography were used to study the inhibitory effect of bitter gourd chitinase on fungi. The results showed that the bitter gourd chitinase could decompose mycelial cell wall preparations of Fusarium oxysporum f. The elongation of a certain inhibition.

目的为研究抗纤复方药物的抗肝纤维化的细胞机制。方法免疫细胞化学染色法比较观察了原代培养正常和纤维肝肝细胞I、III、IV型胶原表达,并以血清药理学方法研究抗纤复方药物对纤维肝肝细胞生成胶原的影响。结果正常肝细胞极少表达胶原,纤维肝肝细胞胶原表达增加,抗纤复方药物能有效抑制纤维肝的肝细胞生成胶原,其作用优于秋水仙碱。结论抗纤复方药物能抑制纤维肝肝细胞合成胶原,而减少肝脏细胞外基质的沉积。
