目的:检测与乳腺癌转移特性相关的雄激素受体(AR)、细胞周期蛋白D1(CC-ND1)和雌激素受体2(ESR2)基因表达,建立与乳腺癌术后复发转移相关的基因表达谱,初步探讨其与乳腺癌复发转移的关系。方法:81例乳腺癌患者术后5年随访期内出现复发者为研究组(41例),未复发仍生存者为对照组(40例)。提取所有病例石蜡包埋组织的RNA,质量鉴定后,应用RT-PCR芯片技术平行检测与乳腺癌转移特性相关的AR、CCND1和ESR2基因,分析基因表达差异与乳腺癌复发之间的关系。结果:AR、CC-ND1和ESR2基因表达水平在复发与未复发组间差异有统计学意义,P值均<0.05。AR和CCND1基因在复发病例中均高表达(62.5%),而ESR2基因在复发组中呈低表达(63.4%)。在ER阳性乳腺癌病例中,CCND1和ESR2基因表达与疾病复发和复发时间均相关,P<0.05。结论:乳腺癌术后AR、CC-ND1和ESR2基因的差异表达与复发转移相关,可做为预防和控制该病复发的新线索。
OBJECTIVE: To detect the expression of androgen receptor (AR), cyclin D1 (CC-ND1) and estrogen receptor 2 (ESR2) genes related to the metastatic characteristics of breast cancer and to establish the genes related to the recurrence and metastasis of breast cancer Expression profiles, preliminary study of its relationship with recurrence and metastasis of breast cancer. Methods: Eighty-one breast cancer patients were recruited as study group (41 cases) and non-recurrent survivors as control group (40 cases) after 5 years of follow-up. The RNA of paraffin-embedded tissue was extracted from all the cases. After quality identification, AR, CCND1 and ESR2 genes related to metastasis of breast cancer were detected by RT-PCR chip in parallel. The relationship between gene expression and recurrence of breast cancer was analyzed. Results: There was significant difference in the expression of AR, CC-ND1 and ESR2 between recurrence and non-recurrence (P <0.05). Both AR and CCND1 were highly expressed in relapsed cases (62.5%), while ESR2 gene was low (63.4%) in relapsed cases. In cases of ER-positive breast cancer, CCND1 and ESR2 gene expression correlated with disease recurrence and relapse time, P <0.05. Conclusion: The differential expression of AR, CC-ND1 and ESR2 genes after breast cancer surgery is related to recurrence and metastasis, which can be used as a new clue to prevent and control recurrence of the disease.
