目的炎症通过其对肿瘤微环境的影响在癌症的发展过程中起着重要作用。然而炎症与肿瘤之间的关系仍然需要进一步阐明。本研究主要观察IL-21R在舌鳞状细胞癌的表达和分布情况。方法使用逆转录酶聚合酶链反应(RT-PCR)、Western blot免疫印迹方法检测来自54个舌鳞状细胞癌患者手术标本IL-21R的基因和蛋白表达,并且观察IL-21R表达与临床病理特征的关系。同时,进一步通过免疫组织化学和免疫荧光技术观察IL-21R表达与分布规律。结果 RT-PCR和Western印迹分析的结果表明,与癌旁组织相比,舌鳞状细胞癌标本中IL-21R基因和蛋白表达显著降低(P<0.05)。IL-21R低表达与性别、年龄、分化程度、肿瘤大小均无密切关系。这种表达特点主要与临床分期呈正相关。免疫组织化学结果显示,IL-21R主要表达于舌癌旁组织鳞状细胞和上皮细胞,而鳞状细胞癌组织中表达较弱或散在的表达。IL-21R与IL-10的双染荧光结果显示,IL-21R阳性的细胞中可见IL-10的阳性染色。结论这些数据表明IL-21R在舌癌肿瘤发生与侵袭性过程中可能起着重要作用。
Aim Inflammation plays an important role in the development of cancer through its effects on the tumor microenvironment. However, the relationship between inflammation and cancer still needs to be further elucidated. This study mainly observed the expression and distribution of IL-21R in tongue squamous cell carcinoma. Methods RT-PCR and Western blot were used to detect the gene and protein expression of IL-21R in surgical specimens of 54 patients with squamous cell carcinoma of the tongue. Immunohistochemical results showed that IL-21R was mainly expressed in squamous cells and epithelial cells of tongue adjacent tissues, while squamous cell carcinoma expressed weakly or scatteredly. Double-staining of IL-21R with IL-10 showed positive staining of IL-10 in IL-21R positive cells. Conclusions These data indicate that IL-21R may play an important role in the carcinogenesis and invasiveness of tongue cancer.
