目的掌握山东口岸鼠类种类以及携带汉坦病毒的情况。方法 2016年在山东滨州、董家口、东营、黄岛、莱州、青岛、青岛机场、威海等口岸用夹夜法捕获鼠类,进行形态学鉴定。从鼠尾中提取线粒体DNA,用通用引物扩增细胞色素C氧化酶亚基I(CO I)基因进行分子鉴定。用实时荧光PCR法检测汉坦病毒。结果共捕获4种76只鼠类,全部扩增到CO I基因,序列比对结果与形态学鉴定相同。褐家鼠和黑线姬鼠的汉坦病毒检测阳性率分别为6.67%和11.11%,小家鼠和小麝鼩未检测到汉坦病毒。结论褐家鼠和黑线姬鼠是山东口岸常见鼠类,均携带汉坦病毒,应加强山东口岸鼠类监测。
Objective To understand the species of rodent in Shandong port and carry Hantaan virus. Methods In 2016, mice were harvested at night in Binzhou, Dongjiakou, Dongying, Huangdao, Laizhou, Qingdao, Qingdao Airport and Weihai by morphological identification. Mitochondrial DNA was extracted from the tail of the rat and the universal primer was used to amplify the cytochrome C oxidase subunit I (COI) gene for molecular identification. Detection of Hantavirus by Real - time Fluorescent PCR. Results A total of 76 murine species were captured and all of them were amplified to CO I gene. The results of sequence alignment were the same as morphological identification. The positive rates of Hantavirus in Rattus norvegicus and Apodemus agrarius were 6.67% and 11.11%, respectively. Hantavirus was not detected in Mus musculus and Muskrat. Conclusion Both Rattus norvegicus and Apodemus agrarius are common rodents in Shandong port, all harboring Hantavirus, so rodent surveillance in Shandong port should be strengthened.
