目的分析乌鲁木齐地区哈萨克族献血人群HPA 1-17系统基因多态性分布特点。方法采用PCR-SSP对乌鲁木齐地区无血缘关系的哈萨克族献血人群100例血液标本进行HPA1-17系统基因分型,计算基因型频率和基因频率。结果哈萨克族献血人群HPA的基因频率分别为:HPA-1a 0.995、-1b 0.005、-2a 0.95、-2b 0.05、-3a 0.52、-3b 0.48、-4a 0.995、-4b 0.005、-5a 0.99、-5b 0.01、-6a 0.99、-6b 0.01、-15a 0.475、-15b 0.525;HPA-3和15系统表现出较高的杂合度,其中3a/3a、3a/3b、3b/3b基因型频率分别为0.3、0.44、0.26;15a/15a、15a/15b、15b/15b基因型频率分别为0.21、0.53、0.26;HPA-7-14、16、17系统只检出a基因,呈单特异性。结论乌鲁木齐地区哈萨克族HPA基因分布具有自身特点,数据分布符合Hardy-Weinberg遗传平衡定律,其中HPA-3和HPA-15抗原不配合率较高。
Objective To analyze the distribution of HPA 1-17 gene polymorphism in Kazakh blood donors in Urumqi. Methods PCR-SSP was used to genotype HPA1-17 gene in 100 blood samples of blood donors without blood relationship in Urumqi region to calculate genotype frequency and gene frequency. Results The gene frequencies of HPA in blood donors from Kazak were: HPA-1a 0.995, -1b 0.005, -2a 0.95, -2b 0.05, -3a 0.52, -3b 0.48, -4a 0.995, -4b 0.005, -5a 0.99, 5b 0.01, -6a 0.99, -6b 0.01, -15a 0.475, -15b 0.525, respectively. The HPA-3 and 15 systems showed higher heterozygosity. The genotype frequencies of 3a / 3a, 3a / 3b and 3b / 3b were 0.3, 0.44, 0.26; 15a / 15a, 15a / 15b, 15b / 15b genotype frequencies were 0.21,0.53,0.26; HPA-7-14,16,17 system only detected a gene was monospecific. Conclusion The Kazak HPA gene distribution in Urumqi has its own characteristics. The data distribution accords with the Hardy-Weinberg law of genetic balance, in which HPA-3 and HPA-15 antigen do not mix well.
