目的 研究二氢毗陡类钙桔抗剂,[2,6 二甲基-4-(3-硝基苯基)-1,4-二氢-3,5-吡啶二羧酸-3-甲酯-5-正戊酯(Mn9202)了、拉西地平(lacidipine)对培养的大鼠胃底平滑肌 5-HT、5-HT合成酶(TH)及 5HT受体(5-HTR)的影响。方法采用免疫细胞化学和图像分析等方法观察Mn9202和lacidipine作用下培养的大鼠胃底平滑肌细胞中5-HT、TH和5-HTR含量的变化.并与5-HT的桔抗剂赛庚定(cyprohetadine)比较。结果 正常大鼠胃底平滑肌细胞5-HT、TH、5-HTR免疫反应均呈阳性,而Mn9202、lacidpine和cyprohetadine均能使胃底平滑肌5-HT、TH和5-HTR的含量减少。结论 大鼠胃底平滑肌胞具有自分泌5-HT的功能,Mn9202和拉西地平可能通过减少5-HT的合成而使5-HTR的表达下降,这一结果与5-HT桔抗剂赛庚定相似。
OBJECTIVE To study the inhibitory effects of dihydropyropene calcium antagonist, [2,6-dimethyl-4- (3-nitrophenyl) -1,4-dihydro-3,5-pyridinedicarboxylate 5-n-pentyl ester (Mn9202), and lacidipine on 5-HT, 5-HT synthase (TH) and 5HT receptor (5-HTR) in cultured rat gastric fundus. Methods Immunocytochemistry and image analysis were used to observe the changes of 5-HT, TH and 5-HTR in rat gastric fundus smooth muscle cells cultured with Mn9202 and lacidipine. And compared with the 5-HT orange antagonist cyprohetadine. Results The 5-HT, TH and 5-HTR immune responses in normal rat gastric fundus smooth muscle cells were all positive. However, Mn9202, lacidpine and cyprohetadine all reduced the content of 5-HT, TH and 5-HTR in gastric fundus. Conclusions Rat gastric fundus smooth muscle cells have the function of autocrine 5-HT. Mn9202 and lacidipine may reduce the expression of 5-HTR by reducing the synthesis of 5-HT. This result is consistent with that of 5-HT, Set similar.
