目的建立瓜子中酸性橙Ⅱ的高效液相色谱(HPLC)测定方法。方法选择HPLC法测定瓜子中酸性橙Ⅱ的理想色谱条件,并测定瓜子中酸性橙Ⅱ的含量及加标回收率,作方法学论证。结果酸性橙Ⅱ在0.1~20μg/ml的范围内线性良好,线性方程为y=34.94x-2.697,r=0.999 8。酸性橙Ⅱ最低检出浓度为0.08 mg/L。样品添加浓度为0.5~10 mg/kg时,方法回收率为51.6%~57.5%。结论以HPLC法检测非法添加于瓜子中酸性橙Ⅱ的含量,操作简便,定量准确可靠。
Objective To establish a method for the determination of acid orange Ⅱ in melon seeds by high performance liquid chromatography (HPLC). Methods The HPLC method was used to determine the ideal chromatographic conditions of acidic orange Ⅱ in melon seeds. The content of acid orange Ⅱ in seeds and the recoveries were determined. Results Acid Orange Ⅱ showed good linearity in the range of 0.1 ~ 20μg / ml. The linear equation was y = 34.94x-2.697, r = 0.999 8. Acid Orange Ⅱ minimum detectable concentration of 0.08 mg / L. When the sample concentration was 0.5 ~ 10 mg / kg, the recovery rate was 51.6% ~ 57.5%. Conclusion The content of acid orange Ⅱ illegally added to melon seeds was detected by HPLC. The method was simple, accurate and reliable.
