目的探讨大鼠脊髓源神经干细胞与运动神经元的差异蛋白质组学,寻找出重要的差异蛋白质。方法采用双向电泳分离两种细胞的蛋白质,用DeCyder软件分析蛋白表达差异,并采用质谱(HPLC-ESI-MS/MS)进行鉴定。结果脊髓源神经干细胞与运动神经元蛋白质凝胶分析获得1 300余个清晰的蛋白质斑点,在比国产分析的87个差异点中,初步鉴定出44个差异表达蛋白,其中24个在神经干细胞高表达,20个在运动神经元高表达。结论脊髓源神经干细胞与运动神经元有各自特定的蛋白质表达,某些差异蛋白可能在神经干细胞向运动神经元分化中发挥关键作用。
Objective To explore the differential proteomics of rat spinal cord-derived neural stem cells and motor neurons and to find important differentially expressed proteins. Methods Two-dimensional electrophoresis (SDS-PAGE) was used to separate the proteins of the two cells. The protein expression differences were analyzed by DeCyder software and identified by mass spectrometry (HPLC-ESI-MS / MS). Results A total of more than 1 300 clear protein spots were obtained from protein gel analysis of spinal cord-derived neural stem cells and motor neurons. Of the 87 differences from the domestic analysis, 44 differentially expressed proteins were identified, 24 of which were highly expressed in neural stem cells Expression, high expression of 20 in motor neurons. CONCLUSIONS: Spinal cord-derived neural stem cells and motor neurons have their own specific protein expression. Some differentially expressed proteins may play key roles in the differentiation of neural stem cells into motor neurons.
