目的:探讨复方中药抗肿瘤的机制及作用靶点。方法:造模后,荷瘤鼠随机分成6组,分别给生理盐水(空白组)、养阴清热方——安体优Ⅰ号低(ATU-L)、中(ATU-M)、高(ATU-H)3个剂量组灌胃、及阳性对照α-IFN和紫杉醇(PTX)组治疗,第14、21天处死动物,制备相应标本,以聚丙烯酰胺凝胶电泳—银染法及TRAP-PCR-ELISA法,定性、定量测定端粒酶活性。结果ATU-H组及ATU-M组对荷瘤小鼠的端粒酶活性有一定抑制作用,在治疗第14天及第21天,ATU-H组端粒酶活性均为阴性,ATU-L组均为阳性,而ATU-M组在第14天为阳性,在第21天为阴性,电泳银染法也显示了相近结果。结论:安体优Ⅰ号对615小鼠HCa-F肝癌的端粒酶活性具有一定的抑制作用。
Objective: To investigate the anti-tumor mechanism and target of compound Chinese medicine. Methods: After the model was established, the tumor-bearing mice were randomly divided into 6 groups: saline (blank group), ATU-L, ATU- ATU-H) were intragastrically administrated in 3 groups, and the positive control group was treated with α-IFN and paclitaxel (PTX). Animals were sacrificed on the 14th and 21st day, and the corresponding samples were prepared. Polyacrylamide gel electrophoresis- -PCR-ELISA, qualitative and quantitative determination of telomerase activity. Results ATU-H group and ATU-M group inhibited the telomerase activity of tumor-bearing mice. On the 14th and 21st days of treatment, the telomerase activity of ATU-H group was negative, ATU-L Group were positive, while the ATU-M group was positive on the 14th day, negative on the 21st day, electrophoretic silver staining also showed similar results. CONCLUSION: The effect of Antaiyou Ⅰ on the telomerase activity of HCa-F hepatocarcinoma in 615 mice was inhibited.
