目的 :观察吡柔比星 ( pirarubicin ,THP)对结肠癌细胞株SW 2 6 0凋亡的影响。 方法 :将THP加入SW 2 6 0细胞培养液 ,Hoechst 3 3 2 5 8荧光染色、透射电镜观察SW 2 6 0细胞形态学变化 ;DNA抽提琼脂糖电泳观察DNA的“梯状”条带 ;并测定细胞DNA断裂百分率 ,以探讨THP能否引起人结肠癌细胞株SW 2 6 0发生凋亡。结果 :不同浓度THP ( 5 ,10 ,15 μmol/L)作用于SW 2 6 0细胞 2 4小时及 10 μmol/LTHP作用于SW 2 6 0细胞不同时间 ( 12 ,2 4,48小时 )后 ,Hoechst 3 3 2 5 8染色荧光显微镜观察及透射电镜观察均可见细胞出现典型凋亡形态学改变 ;DNA抽提琼脂糖电泳出现凋亡细胞特征性生化变化 DNA“梯状”条带 ;DNA断裂百分率则随着THP浓度的增大或作用时间延长而逐渐增高。结论 :THP能诱导体外培养的结肠癌细胞株SW 2 6 0发生凋亡
Objective: To observe the effect of pirarubicin (THP) on the apoptosis of colon cancer cell line SW 2 600. METHODS: THP was added to the culture medium of SW 2 600 cells. Hoechst 3 3 258 fluorescence staining and transmission electron microscopy were used to observe the morphological changes of SW 260 cells. DNA extraction agarose electrophoresis was used to observe the “ladder” bands of DNA. The percentage of cell DNA fragmentation was measured to determine whether THP can cause apoptosis of human colon cancer cell line SW260. Results: Different concentrations of THP (5, 10, 15 μmol/L) were applied to SW 2 600 cells for 24 hours and 10 μmol/LTHP were applied to SW 2 600 cells at different times (12, 24 hours). The morphology of typical apoptotic cells was observed by Hoechst 3 3 2 5 8 staining and transmission electron microscopy. The characteristic “a ladder-like” bands of apoptotic cells were detected by DNA extraction agarose gel electrophoresis; DNA fragmentation percentage was observed. With the increase of THP concentration or prolonged action time, it gradually increased. Conclusion :THP can induce the apoptosis of colon cancer cell line SW 260 in vitro
