以细胞培养法并用肝素等作对照,观察类肝素对兔主动脉血管平滑肌细胞(VSMCs)增生的影响。结果表明,类肝素在16~005mg·ml-1浓度下,对FCS,bFGF,IL1刺激细胞增殖均有明显的抑制作用。将VSMCs在类肝素02~005mg·ml-1中预孵24h后弃去药液,再在含FCS培养液中继续培养,也能明显抑制细胞增殖。类肝素对FCS及IL1诱导VSMCs增殖的抑制作用比同浓度的肝素弱,但对bFGF诱导VSMCs增殖的抑制作用比肝素强。右旋糖酐对VSMCs增殖无抑制作用,提示抑制作用与分子中含硫酸基有关。
The effects of heparins on proliferation of rabbit aortic vascular smooth muscle cells (VSMCs) were observed by cell culture and heparin. The results showed that heparin at 1 6 ~ 0 05mg · ml-1 concentrations, FCS, bFGF, IL 1 stimulated cell proliferation were significantly inhibited. The VSMCs were preincubated for 24 h in heparinoid 0 2 ~ 0 05 mg · ml-1 and then discarded after further incubation in FCS-containing medium, but also significantly inhibited cell proliferation. Compared with the same concentration of heparin, the inhibitory effect of heparin on FCS and IL-1 induced VSMCs proliferation was weaker than that of heparin on bFGF-induced proliferation of VSMCs. Dextran had no inhibitory effect on VSMCs proliferation, suggesting that the inhibitory effect was related to the presence of sulfate groups in the molecule.
