目的:探讨参麦总皂苷抗血管生成作用,为临床辅助抗肿瘤分子机理提供理论依据。方法:提取参麦总皂苷,采用MTT法及刮痕法分析总皂苷对人脐静脉内皮细胞HUVECs的增殖和迁移抑制作用;通过荧光定量PCR方法考察总皂苷对人肝癌HepG2细胞株VEGF表达影响;最后,以鸡胚绒毛尿囊膜(CAM)在体模型对总皂苷抗血管生成作用进行评价。结果:MTT及刮痕实验结果表明,参麦总皂苷可剂量依赖性抑制HUVECs细胞增殖和迁移,其中600μg/mL浓度增殖抑制率达63.9%,150μg/mL处理48h,其迁移抑制率可达73.7%;荧光定量PCR结果表明,总皂苷可以抑制HepG2细胞分泌VEGF;此外,鸡胚绒毛尿囊膜模型证实,参麦总皂苷具有抗血管生成作用。结论:参麦总皂苷可以通过抑制内皮细胞增殖、迁移及抑制癌细胞分泌VEGF表达起到抗肿瘤血管生成作用。
Objective: To explore the anti-angiogenesis effect of total saponins of Shenmai and to provide a theoretical basis for clinical molecular mechanism of anti-tumor. Methods: The total saponin of Shenmai was extracted, the proliferation and migration of HUVECs were analyzed by MTT assay and scratch assay. The effect of total saponins on the expression of VEGF in human hepatocellular carcinoma HepG2 cells was investigated by fluorescence quantitative PCR. Finally, the anti-angiogenic effect of total saponins in chick chorioallantoic membrane (CAM) was evaluated in vivo. Results: The results of MTT and scratch test showed that the total saponin of SSP could inhibit the proliferation and migration of HUVECs in a dose-dependent manner. The inhibitory rate of proliferation was 63.9% at 600μg / mL, 48h at 150μg / mL, and the inhibition rate was 73.7 %. The result of real-time PCR showed that total saponins could inhibit the secretion of VEGF in HepG2 cells. In addition, chick chorioallantoic membrane model confirmed that total saponins of Shenmai had anti-angiogenic effect. Conclusion: SSP can play an anti-angiogenic effect by inhibiting the proliferation and migration of endothelial cells and inhibiting the expression of VEGF secreted by cancer cells.
