目的探讨肝细胞癌组织结肠癌转移相关基因1(metastasis-associated in colon cancer 1,MACC1)的表达及其临床意义。方法采用免疫组化测定108例病理确诊肝细胞癌患者的肝癌组织和癌旁无瘤肝组织中的MACC1蛋白表达情况,实时荧光定量PCR检测30例肝癌组织、癌旁组织以及20例正常肝组织中MACC1 mRNA的表达,分析肝癌组织中MACC1的表达水平与临床病理因素及预后的关系。结果免疫组化结果显示,肝癌组织中MACC1蛋白表达的阳性率为49.1%(53/108),高于癌旁肝组织34.2%(37/108,P=0.011);实时荧光定量PCR结果显示,MACC1mRNA在肝癌组织中的表达高于癌旁肝组织及正常肝组织(P<0.001)。单因素方差分析表明肝癌组织中MACC1的表达与肿瘤大小、有无包膜及病理分型密切相关(P<0.05)。生存分析表明MACC1阳性患者的1年、3年和5年总体生存率分别为0.650、0.492和0.280,而MACC1阴性患者1年、3年和5年总体生存率分别0.799、0.684和0.566,差异有统计学意义(P=0.006)。结论肝癌组织中MACC1的表达与肝癌的恶性演变及患者的临床预后密切相关,MACC1有望成为判断肝癌患者临床预后的重要分子,且有可能成为肝癌分子靶向治疗的新靶点。
Objective To investigate the expression of metastasis-associated in colon cancer 1 (MACC1) in hepatocellular carcinoma (HCC) and its clinical significance. Methods Immunohistochemistry was used to detect the expression of MACC1 protein in 108 hepatocellular carcinoma patients with pathologically confirmed hepatocellular carcinoma (HCC) and adjacent non-cancerous liver tissues. Real-time fluorescence quantitative PCR was used to detect the expression of MACC1 in 30 HCC tissues, adjacent normal tissues and 20 normal liver tissues MACC1 mRNA expression in liver cancer tissue MACC1 expression levels and clinicopathological factors and prognosis. Results The positive rate of MACC1 protein in hepatocellular carcinoma was 49.1% (53/108), higher than that in adjacent noncancerous liver tissue (34.2%, 37/108, P = 0.011). The results of real-time fluorescence quantitative PCR showed that, The expression of MACC1mRNA in HCC tissue was higher than that in paracancer liver tissue and normal liver tissue (P <0.001). One-way ANOVA showed that the expression of MACC1 in HCC was closely related to the size of the tumor, the presence or absence of the capsule and pathological classification (P <0.05). Survival analysis showed that the 1-year, 3-year and 5-year overall survival rates of MACC1-positive patients were 0.650,0.492 and 0.280 respectively, while the overall survival rates of MACC1-negative patients at 1 year, 3 years and 5 years were 0.799, 0.684 and 0.566 respectively Statistical significance (P = 0.006). Conclusion The expression of MACC1 in hepatocellular carcinoma is closely related to the malignant transformation of hepatocellular carcinoma and the clinical prognosis of patients with hepatocellular carcinoma. MACC1 is expected to be an important molecular prognostic marker in patients with hepatocellular carcinoma and may become a new target of molecular targeted therapy for hepatocellular carcinoma.
