目的:研究SALL4基因在宫颈癌、宫颈上皮内瘤变(cervical intraepithelial neoplasia,CIN)组织和血浆中的表达情况并探讨其意义。方法:采用实时定量PCR方法检测56例宫颈癌患者、120例CIN患者组织和血浆中SALL4 mRNA的表达,以35例正常宫颈及慢性宫颈炎组织和血浆作为对照,分析宫颈癌组织和血浆中SALL4 mRNA相对表达量的相关性。并用ELISA方法检测各组血浆中SALL4水平。结果:对照组、CINⅠ组、CINⅡ~Ⅲ组、宫颈癌组组织中SALL4 mRNA相对表达水平分别为(4.58±0.64)×10-4、(6.86±0.72)×10-4、(27.03±2.85)×10-4、(40.8±5.50)×10-4,血浆中SALL4 mRNA相对表达水平分别为(0.14±0.05)×10-4、(0.16±0.07)×10-4、(0.49±0.08)×10-4、(2.18±0.22)×10-4,血浆中SALL4的浓度分别为(237±45)μg/L、(256±59)μg/L、(713±62)μg/L、(1 135±107)μg/L,随着病变的逐步进展,SALL4的表达逐渐增强,其中SALL4在宫颈癌组中的表达显著增强,与其他各组比较,差异有统计学意义(P<0.01).宫颈癌组组织和血浆中SALL4 mRNA的相对表达水平正相关(r=0.684,P<0.01)。结论:SALL4基因在CIN、宫颈癌组织和血浆中表达上调,可能在宫颈癌发生、发展中起一定作用。
Objective: To investigate the expression of SALL4 gene in cervical cancer, cervical intraepithelial neoplasia (CIN) tissues and plasma and to explore its significance. Methods: The mRNA and protein expression of SALL4 in 56 cases of cervical cancer and 120 cases of CIN were detected by real-time quantitative polymerase chain reaction (RT-PCR). 35 cases of normal cervix and chronic cervicitis and plasma were used as controls to analyze the expression of SALL4 Correlation of mRNA relative expression. The levels of SALL4 in plasma were detected by ELISA. Results: The relative expression levels of SALL4 mRNA in control group, CINⅠgroup, CINⅡ ~ Ⅲ group and cervical cancer group were (4.58 ± 0.64) × 10-4, (6.86 ± 0.72) × 10-4, (27.03 ± 2.85) × 10-4 and (40.8 ± 5.50) × 10-4 respectively. The relative expression levels of SALL4 mRNA in plasma were (0.14 ± 0.05) × 10-4, (0.16 ± 0.07) × 10-4, (0.49 ± 0.08) × The plasma SALL4 concentrations were (237 ± 45) μg / L, (256 ± 59) μg / L, (713 ± 62) μg / L, 135 ± 107) μg / L. The expression of SALL4 was gradually increased with the progression of the lesions. The expression of SALL4 in cervical cancer group was significantly higher than that in other groups (P <0.01). There was a positive correlation between the relative expression of SALL4 mRNA in cervical cancer tissues and plasma (r = 0.684, P <0.01). Conclusion: SALL4 gene is upregulated in CIN, cervical cancer and plasma, which may play a role in the occurrence and development of cervical cancer.
