系统地研究了诱导高频、同步可制成“芽珠”的赤桉幼芽(Shoottips)的方法,为以幼芽为繁殖体制备人工种子奠定基础。试验结果:首先,培养基中适宜的BA和NAA浓度和配比直接控制着制备“芽珠”型幼芽的数量,同时对已分化出的幼芽进一步伸长有很大的影响,BA为1.0mg/L,NAA为0.5mg/L的培养基中附加40mg/L腺嘌呤和5%的蔗糖能较好地控制幼芽生长及其叶片大小;其次,培养过程中先以16天光/暗为10h/14h培养之后转入4天全黑暗培养可控制幼芽生长的同步性,在半固体培养基上连续继代三次,获得了每次继代(20天)平均产生25个/cm2适宜制备“芽珠”的幼芽。文中就赤桉培养中褐色渗出物问题进行了初步研究,通过预培养的方法克服了褐变物对培养中材料的影响。
The method of inducing high frequency and synchronizing Shoottips which can be made into “bud beads” was systematically studied, which lays the foundation for preparing artificial seeds with shoots as propagules. Test results: Firstly, the appropriate concentration of BA and NAA in the medium and the ratio directly controlled the number of sprouting buds, and at the same time had a great influence on the further elongation of buds that had already been differentiated. BA was 1.0mg / L, NAA 0.5mg / L medium supplemented with 40mg / L adenine and 5% sucrose can better control the bud growth and leaf size; secondly, the culture process to 16 days of light / Dark for 10h / 14h transferred to four days after dark culture can control the growth of all dark synchronization of the growth of young shoots, semi-solid medium for three times in succession, obtained each generation (20 days) an average of 25 / cm2 suitable for the preparation of “bud beads” buds. In the paper, the brown exudate in the cultivation of Eucalyptus globulus was preliminarily studied, and the influence of browning on the materials in culture was overcome by the method of pre-culture.
