目的论关于眶额叶区5-HT以及Glu表达水平与脑梗死后抑郁症关联。方法健康成年雄性大鼠进行实验,饲养1周后造模,造模组动物6只;同时选取对照6只,进行实验研究。借助高效液相色谱实验进行5-HT和多巴胺的含量测定;Glu含量检测首先制备标准品后制备样品,进样条件:流速:1.0 m L/min,检测波长为360 nm。实验结束后,麻醉乌拉坦麻醉实验动物后微量注射澳酚兰溶液后处死取脑,多聚甲醛固定,冰冻切片机作连续的冠状切片,镜检染液的组织学部位。统计分析采用SPSS19.0统计软件包完成,结果效液相色谱分析,测定5-HT标准品溶液峰面积,出峰时间为7.367 min,绘制标准曲线,其线性范围为2~300 ng/m L;建模组与对照组5-HT含量测定结果提示,造模组5-HT表达水平为(341.89±36.98)ng/g,对照组为(335.77±50.89)ng/g,两组水平未见显著差别(P>0.05)。进一步测定不同浓度下的Glu标准品溶液峰面积,出峰时间为11.231 min,线性范围14.1~142μg/m L;各组Glu含量存在差别,造模组高于对照组水平,两组Glu水平分别为(1352.34±431.22)μg/g和(976.25±85.66)μg/g,(P<0.01)。最后,造模组与对照组5-HT表达未见明显差别,继续通过给与外源性注射5-HT 1A受体激动剂8-OH-DPAT后观察,发现大鼠的抑郁样行为得到显著改善,主要表现为糖水偏爱率的提高等方面。结论脑卒总后抑郁样行为的发生可能与眶额叶5-HT水平存在关联,但并不因其直接引发,而可能是通过Glu能系统的介导后产生的,该调节作用由1A受体介导,Glu能系统异常会导致5-HT抗抑郁的效果受影响或无法实现。

Six weeks after the feeding, six rabbits were sacrificed and six rabbits in the model group were selected for experiment.
