目的探讨膀胱癌的端粒酶活性表达及其在膀胱癌中的诊断价值。方法采用端粒重复序列扩增程序──酶标法(定量)和银染法(定性)对28例膀胱瘤、11例癌旁膀胱、6例正常膀胱的组织标本进行端粒酶活性检测。结果膀胱癌组织的端粒酶活性阳性检出率为89.3%(25/28),癌旁膀胱组织为27.3%(3/11),正常膀胱组织为0%(0/6)。膀胱癌组与癌旁组、正常组端粒酶阳性检出率经比较,有显著性差异(P<0.01)。端粒酶活性表达与膀胱癌病理分期、分级等无相关性(P>0.05)。结论端粒酶激活发生在肿瘤形成的早期阶段,有望成为检测早期初发或复发膀胱癌有用的肿瘤标志物,癌旁组织端粒酶的激活提示肿瘤微浸润可能。
Objective To investigate the expression of telomerase activity in bladder cancer and its diagnostic value in bladder cancer. Methods telomerase activity was detected in 28 cases of bladder tumor, 11 cases of paracancerous bladder and 6 cases of normal bladder tissue by telomeric repeat amplification protocol (ELISA) (quantitative) and silver staining (qualitative). Results The positive rate of telomerase activity was 89.3% (25/28) in bladder cancer tissues, 27.3% (3/11) in para-cancerous tissues and 0% (0/6) in normal bladder tissues. . The positive rate of telomerase in bladder cancer group, adjacent normal group and normal group was significantly different (P <0.01). There was no correlation between telomerase activity and pathological stage and grade of bladder cancer (P> 0.05). Conclusion Telomerase activation takes place in the early stage of tumor formation and is expected to be a useful tumor marker for the detection of early or recurrent bladder cancer. The activation of telomerase in para-cancerous tissues suggests that micro-infiltration of tumor may be possible.
