目的探讨新疆软紫草有效成分紫草素对D半乳糖所致老年性痴呆模型大鼠学习记忆功能的影响。方法雄性Wistar大鼠腹腔注射D半乳糖生理盐水(150mg/kg.d)连续6周,继以紫草素高(0.7mg/100g.d)、低(0.35mg/100g.d)剂量灌胃给药1周。以Morris水迷宫测定大鼠空间学习记忆能力、以黄嘌呤氧化酶法和硫代巴比妥酸法分别测定大鼠脑匀浆中超氧化物歧化酶(superoxide dis-mutase,SOD)活力及丙二醛(maleic dialdehyde,MDA)含量。结果与对照组相比,模型组的学习记忆能力和SOD活力明显降低、MDA含量明显升高(P<0.05,P<0.01),紫草素高剂量组具有一定的改善AD模型大鼠学习记忆的作用,并可明显提高模型大鼠脑匀浆中的SOD活力、降低其中的MDA含量(P<0.05,P<0.01)。结论紫草素可明显提高模型大鼠的学习记忆能力、拮抗脑内氧化胁迫损伤。
Objective To investigate the effect of shikonuen (Compositae purpurea), an active ingredient of Xinjiang soft grass, on the learning and memory abilities of Alzheimer’s disease (AD) model induced by D-galactose in rats. Methods Male Wistar rats were intraperitoneally injected with D-galactose normal saline (150mg / kg.d) for 6 weeks, followed by intragastric administration of shikonin (0.7mg / 100g.d) and low dose (0.35mg / 100g.d) Administered for 1 week. Morris water maze was used to measure the spatial learning and memory abilities in rats. The activities of superoxide dismutase (SOD) and brain tissue were measured by xanthine oxidase and thiobarbituric acid respectively. Maleic dialdehyde (MDA) content. Results Compared with the control group, the learning and memory ability and the activity of SOD in the model group were significantly decreased, and the content of MDA was significantly increased (P <0.05, P <0.01). The shikonin high dose group had certain learning and memory abilities (P <0.05, P <0.01), and significantly increased the SOD activity in brain homogenates of model rats. Conclusion Shikonin can significantly improve the learning and memory ability of model rats and antagonize the oxidative stress injury in the brain.
