[目的]研究胃癌组织中第10号染色体同源丢失性磷酸酶-张力蛋白基因(PTEN)与多种肿瘤抑制基因(P16)、血管内皮生长因子(VEGF)的表达及其相关性,探讨其与胃癌生物学行为的关系及意义。[方法]应用免疫组化S-P法检测40例胃癌患者胃癌组织和10例正常胃黏膜组织中PTEN、P16、VEGF的表达情况及其与胃癌临床生物学行为的关系。[结果]胃癌组织中PTEN蛋白表达下降,VEGF蛋白表达上升,其表达水平与胃癌患者的性别、年龄及分化程度均无明显相关(P>0.05),而与患者的淋巴结转移、临床病理分级、分化程度和浸润深度有关(P<0.05);通过Spearman秩相关检验发现在胃癌组织中,PTEN与P16的表达呈正相关(P<0.01),均呈下降趋势;PTEN与VEGF的表达及P16与VEGF的表达均呈负相关(P<0.01)。[结论]胃癌组织中PTEN和P16表达下降,VEGF表达上升,与胃癌临床生物学行为显著相关,提示三者表达异常参与了胃癌的发生、发展。
[Objective] To investigate the expression and correlation of PTEN, P16 and VEGF in chromosome 10 in gastric cancer and to investigate the relationship between PTEN and multiple tumor suppressor genes (P16) and vascular endothelial growth factor (VEGF) Relationship with biological behavior of gastric cancer and its significance. [Methods] The expression of PTEN, P16 and VEGF in gastric cancer tissues and 10 cases of normal gastric mucosa tissues from 40 cases of gastric cancer were detected by immunohistochemical S-P method and their relationship with the clinical biological behavior of gastric cancer. [Results] The expression of PTEN protein and VEGF protein in gastric cancer tissues were decreased and the expression levels of VEGF protein were not correlated with the gender, age and differentiation of gastric cancer (P> 0.05), but not with the lymph node metastasis, clinicopathological grade, (P <0.05). The Spearman rank correlation test showed that there was a positive correlation between the expression of PTEN and P16 in gastric cancer (P <0.01), the expression of PTEN and VEGF and the expression of P16 and VEGF (P <0.01). [Conclusion] Decreased expressions of PTEN and P16 and increased expression of VEGF in gastric cancer tissues are significantly correlated with the clinical biological behavior of gastric cancer, suggesting that abnormal expression of these three genes is involved in the occurrence and development of gastric cancer.
