目的应用RNA干扰(RNAi)技术沉默Ki67基因,探讨其对裸鼠人肾癌细胞移植瘤细胞生长及凋亡的影响。方法建立裸鼠人肾癌细胞移植瘤模型,随机分组,于肿瘤内分别注射生理盐水0.2 ml(C组)、pSilencer空质粒20μg/只(P组)、pSilencer-Ki67重组质粒20μg/只(PK组),每隔2天注射一次,共7次。取肿瘤测量体积,免疫组化染色检测Ki67的表达,TUNEL法检测细胞凋亡。结果 PK组移植瘤体积明显小于C、P组,移植瘤细胞Ki67表达率也明显降低,肿瘤细胞凋亡率明显增加(P<0.01)。结论应用RNAi技术沉默Ki67基因可以降低人肾癌细胞裸鼠移植瘤Ki67基因表达,同时引起细胞凋亡进而抑制肿瘤的生长。
Objective To silence the Ki67 gene by RNA interference (RNAi) technology and investigate its effect on the growth and apoptosis of xenografted human renal cell carcinoma cells in nude mice. Methods Human renal cancer cell xenografts were established in nude mice and were randomly divided into three groups: saline (0.2 ml), pSilencer (20 μg / p), pSilencer-Ki67 Group), injected once every 2 days for a total of 7 times. The tumor volume was measured, the expression of Ki67 was detected by immunohistochemical staining and the apoptosis was detected by TUNEL method. Results The volume of xenografts in PK group was significantly smaller than that in C and P groups. The expression of Ki67 in xenograft tumor cells was also significantly decreased, and the apoptosis rate of tumor cells was significantly increased (P <0.01). Conclusion The silencing of Ki67 gene by RNAi can reduce the expression of Ki67 gene in human renal cell carcinoma xenografts in nude mice, induce apoptosis and further inhibit tumor growth.
