目的:探讨表皮生长因子受体Ⅲ型突变体(epidermal growth factor receptor variantⅢ,EGFRvⅢ)在人脑成胶质细胞瘤中的表达及其对成胶质细胞瘤A172细胞增殖的影响。方法:应用免疫组织化学染色法检测EGFRvⅢ在60例人脑成胶质细胞瘤及其癌旁正常脑组织中的表达。建立稳定高表达EGFRvⅢ的成胶质细胞瘤A172细胞株,应用细胞计数法、MTT法、细胞克隆形成实验以及裸鼠成瘤实验检测其对成胶质细胞瘤A172细胞增殖的影响。结果:EGFRvⅢ在脑成胶质细胞瘤组织中的阳性表达率明显高于其癌旁正常脑组织(P<0.01);与转染空载体对照组比较,高表达EGFRvⅢ的A172细胞增殖能力明显增加;对照组和高表达EGFRvⅢ组的细胞克隆数分别为(50±10)个和(140±25)个,差异有统计学意义(P<0.01);高表达EGFRvⅢ组的裸鼠皮下移植瘤的体积和质量均高于对照组(P<0.01)。结论:EGFRvⅢ在人脑成胶质细胞瘤组织中高表达,并且可促进成胶质细胞瘤A172细胞的增殖。
Objective: To investigate the expression of epidermal growth factor receptor variant Ⅲ (EGFRv Ⅲ) in human brain glioblastoma and its effect on the proliferation of glioblastoma A172 cells. Methods: Immunohistochemical staining was used to detect the expression of EGFRv Ⅲ in 60 human glioblastoma and its adjacent normal brain tissue. To establish a stable and high expression of EGFRv Ⅲ A162 cell line of glioblastoma. The cell proliferation, proliferation and proliferation of glioblastoma A172 cells were detected by cell counting method, MTT assay, cell clone formation assay and nude mice tumorigenesis assay. Results: The positive expression rate of EGFRv Ⅲ in brain astrocytoma was significantly higher than that in adjacent normal tissues (P <0.01). Compared with the control group transfected with empty vector, the proliferation of EGFRv Ⅲ highly expressing A172 cells was significantly increased (50 ± 10) and (140 ± 25), respectively, in the control group and the EGFRvⅢ-overexpression group (P <0.01). The number of subcutaneous xenografts Volume and quality were higher than the control group (P <0.01). Conclusion: EGFRv Ⅲ is highly expressed in human glioblastoma tissue and can promote the proliferation of A172 cells.
