[目的]探讨经高强度离心运动后热休克蛋白70(HSP70)在骨骼肌细胞中的的表达以及特征性分析。[方法]采用2~6个月的SD大鼠56只,电动炮台,间歇性离心运动训练方案,随机分为对照组、离心训练4周组、离心训练8周组,每组8只,后将对照组及训练8周的大鼠进行高强度的离心运动;取大鼠的脾细胞、外周淋巴细胞,采用蛋白免疫印迹分析HSP70含量变化,采用免疫组织化学法分析骨架蛋白Desmin的变化。[结果]8周间歇性离心训练后HSP70含量显著增加,训练SD大鼠在进行高强度离心运动时,具有适应性,骨架蛋白Desmin丢失量降低,CK显著降低,表明增加的HSP70对骨架蛋白具有保护作用;对照组及离心训练4周组大鼠在高强度离心后表现出显著的应激反应,骨架蛋白Desmin显著缺失,HSP70显著增加。[结论]热休克蛋白70(HSP70)在高强度离心运动后在骨骼肌中蛋白表达有持续性和延迟性,在引起的骨架蛋白损伤中可能起到补充保护的作用机制。
[Objective] To investigate the expression and characterization of heat shock protein 70 (HSP70) in skeletal muscle cells after high-intensity centrifugation. [Methods] Fifty-six SD rats of 2-6 months, electric fort and intermittent eccentric exercise training were randomly divided into control group, centrifuge training for 4 weeks, and centrifugal training for 8 weeks for 8 weeks. The rats in the control group and the training group for 8 weeks were subjected to high-intensity centrifugation. The spleen cells and peripheral lymphocytes of the rats were taken out. The content of HSP70 was analyzed by Western blotting and the changes of the skeletal protein Desmin were analyzed by immunohistochemistry. [Result] The content of HSP70 increased significantly after 8 weeks of intermittent eccentric training. Training SD rats were adaptive when they were subjected to high-intensity eccentric exercise, the loss of Desmin loss of skeletal protein and the decrease of CK were significantly reduced, indicating that the increased HSP70 had After 4 weeks of centrifugation, rats in control group and centrifugation group showed significant stress response after high-intensity centrifugation. The protein Desmin was significantly deleted and HSP70 was significantly increased. [Conclusion] The expression of heat shock protein 70 (HSP70) in skeletal muscle after sustained high-intensity centrifugation is persistent and delayed, which may play a role in the protection of scaffold proteins.
