目的 :探讨大肠癌组织胃泌素与 P5 3蛋白表达的关系及其对肿瘤增殖活性的影响。方法 :放免法检测 32例大肠癌血清及癌组织胃泌素含量。免疫组化标记癌组织及癌近旁黏膜胃泌素、P5 3蛋白PCNA表达。结果 :癌组织胃泌素与 P5 3蛋白表达强度呈正相关 (r=0 .4 6 6 6 ,P<0 .0 1) ,且二者均与肿癌 PCNA表达强度呈正相关 (r=0 .84 83,P<0 .0 0 1;r=0 .5 6 6 8,P<0 .0 2 5 )。二者共同表达时肿瘤PCNA较各自表达时增高。P5 3蛋白表达与血清、癌组织胃泌素水平升高有关。癌近旁黏膜 P5 3蛋白表达组较未表达组 PCNA表达强度增高 (P<0 .0 5 ) ,但与血清胃泌素水平无关。结论 :大肠癌胃泌素、P5 3蛋白独自或共同表达均可提高大肠癌增殖活性 ;癌近旁黏膜增殖状态与 P5 3基因突变有关 ,但与胃泌素表达及血清胃泌素水平无关
Objective: To investigate the relationship between the expression of gastrin and P53 protein in colorectal carcinoma and its effect on tumor proliferation. Methods: Radioimmunoassay was used to detect gastrin levels in 32 cases of colorectal cancer serum and cancer tissue. Immunohistochemical staining of cancer tissue and adjacent mucosal gastrin, P53 protein PCNA expression. Results: There was a positive correlation between the expression of gastrin and the expression of P53 protein in cancer tissues (r = 0.4666, P <0.01), and both were positively correlated with the expression of PCNA in cancer tissues (r = 0. 84 83, P <0. 01; r = 0. 56 6 8, P <0. 025). The expression of tumor PCNA was higher than the expression of both when they were co-expressed. P53 protein expression and serum, cancer tissue gastrin levels. The expression of PCNA in adjacent mucosa of P5 3 protein expression group was higher than that in non-expression group (P <0.05), but not with serum gastrin level. CONCLUSIONS: Colorectal cancer gastrin and P53 protein alone or in combination can increase the proliferation activity of colorectal cancer. The proliferation status of mucosa adjacent to cancer is related to the mutation of P53 gene but not to the expression of gastrin and the level of serum gastrin
