目的探讨柿叶黄酮(PLF)对阿霉素肾病大鼠肾组织纤维连接蛋白(FN)表达及对肾脏免疫病理损伤的影响。方法制备阿霉素肾病鼠模型,采用PLF干预模型鼠,测定模型鼠各阶段24 h尿蛋白变化(于模型制备前、制备后1周、给药3周及给药5周后测定),应用苏木精-伊红(HE)染色法观测肾脏病理形态变化,用免疫组化法观测肾组织中FN表达。结果 PLF能明显降低模型鼠24 h尿蛋白水平(P<0.01)、改善肾脏病理损伤以及抑制肾组织FN表达(P<0.01)等。结论 PLF能通过降低模型鼠肾组织FN表达、改善肾组织纤维化,对模型鼠肾脏起到一定的保护作用。
Objective To investigate the effect of persimmon leaf flavonoids (PLF) on the expression of FN and the pathological changes of kidney in adriamycin-induced nephropathy rats. Methods Adriamycin-induced nephropathy mouse model was established. The 24-hour urinary protein changes in various stages of the model rats (before model preparation, 1 week after preparation, 3 weeks after drug administration and 5 weeks after drug administration) were determined by PLF intervention model. Hematoxylin and eosin (HE) staining was used to observe the renal pathological changes. The expression of FN in renal tissues was detected by immunohistochemistry. Results PLF could significantly decrease 24 h urinary protein (P <0.01), improve renal pathological injury and inhibit FN expression in renal tissue (P <0.01). Conclusion PLF can improve the renal fibrosis by reducing the expression of FN in the kidney of model mice and play a protective role on the model kidney.
