目的:建立快速溶剂萃取(ASE)-高效液相色谱法测定大叶紫珠中的毛蕊花糖苷的方法。方法:采用ASE350快速溶剂萃取系统提取大叶紫珠中的毛蕊花糖苷;以高效液相色谱法测定其含量。色谱柱为Thermo-AQ-C18,流动相为乙腈-0.5%磷酸(17∶83),流速为0.3 m L/min,检测波长为332nm,柱温为35℃,进样量为1μL。结果:毛蕊花糖苷的进样量线性范围为0.013828~0.082968μg(r>0.9998);精密度、稳定性、重复性试验的RSD<2%;加样回收率为98.08%~100.90%,RSD=0.96%。结论:本方法简便、快速、结果准确,可快速提取及测定大叶紫珠中的毛蕊花糖苷。
OBJECTIVE: To establish a rapid solvent extraction (ASE) - HPLC method for the determination of verbascoside in macrophylla. Methods: ASE350 rapid solvent extraction system was used to extract verbascoside in macrophylla and its content was determined by high performance liquid chromatography (HPLC). The column was Thermo-AQ-C18, the mobile phase was acetonitrile-0.5% phosphoric acid (17:83), the flow rate was 0.3 m L / min, the detection wavelength was 332 nm, the column temperature was 35 ℃ and the injection volume was 1μL. Results: The linear range of verbascoside injection was 0.013828 ~ 0.082968μg (r> 0.9998). The RSD of precision, stability and reproducibility was less than 2%. The recoveries were 98.08% ~ 100.90% and the RSD was 0.96 %. Conclusion: The method is simple, rapid and accurate. It can quickly extract and determine verbascoside in the leaves of large-leaved purple beads.
