摘除卵巢的小白鼠,用雌、孕激素处理,并以橄榄油诱发蜕膜。经人工诱发蜕膜后的小白鼠随机分为两组:实验组皮下注射前列腺素E_2(PGE_2),对照组皮下注射生理盐水。结果PGE_2使蜕膜重量显著减轻,且子宫基质细胞向蜕膜细胞的转化受抑制。皮下注射PGE_2还使蜕膜cAMP含量及DNA合成减少。提示PGE_2可能通过对cAMP的影响和干扰蜕膜组织的核酸代谢,从而使子宫蜕膜细胞反应受抑制。我们认为PGE_2的这种作用是不依赖于卵巢和胚泡的,这可能是PGE_2终止早孕的机理之一。
Ovariectomized mice were treated with estrogen and progesterone and decidualized with olive oil. The decidualized mice were randomly divided into two groups: experimental group subcutaneous injection of prostaglandin E_2 (PGE_2), control group subcutaneous injection of saline. Results PGE 2 significantly reduced the decidual weight, and the conversion of uterine stromal cells to decidual cells was inhibited. Subcutaneous injection of PGE 2 also reduced decidual cAMP content and DNA synthesis. These results suggest that PGE 2 may inhibit uterine decidual cell response by affecting cAMP and interfering with the nucleic acid metabolism of decidual tissue. We believe that this effect of PGE_2 is independent of the ovary and blastocyst, which may be one of the mechanisms of PGE_2 termination of early pregnancy.
