响应曲面法优化纳滤浓缩延胡索水提液的工艺研究

目的利用响应面分析方法优化延胡索提取液的纳滤浓缩工艺。方法以延胡索乙素截留率为指标,在明确压力、温度、平衡体积等因素影响的基础上,采用Box-Behnken中心组和设计建立数学模型,考察截留分子量、浓度、p H对延胡索乙素截留率的影响。结果优选出最佳纳滤浓缩条件为:纳滤膜截留分子量450 Da,药液浓度192.68μg/m L,p H 4.72,延胡索乙素理论截留率为95.00%,实测截留率为(93.99±1.29)%。结论该模型可以较好的预测延胡索乙素的截留率,响应面法对中药纳滤浓缩的参数优选合理可行,为热敏性中药成分的纳滤浓缩提供基础。 Objective To optimize the nanofiltration concentration process of Corydalis yanhusuo extract by response surface methodology. Methods Taking Tetrahydropalmatine retention rate as an index and using the Box-Behnken central group and design to establish a mathematical model based on the explicit influence of factors such as pressure, temperature and equilibrium volume, the effects of interception molecular weight, concentration and p H on the retention of tetrahydropalmatine Rate of impact. Results The optimum conditions for the nanofiltration were as follows: the molecular weight of the nanofiltration membrane was 450 Da, the concentration of the nanofiltration membrane was 192.68 μg / mL, the pH was 4.72, the theoretical retention rate of tetrahydropalmatine was 95.00%, and the measured retention rate was (93.99 ± 1.29 )%. Conclusion The model can predict the retention of tetrahydropalmatine better. The response surface methodology is reasonable and feasible for the parameters of nanofiltration and concentration, and provides the basis for the nanofiltration concentration of the components of thermosensitive traditional Chinese medicine.
