目的研究黄芪醇提取物对糖尿病大鼠冠状小动脉平滑肌张力的影响。方法通过ip小剂量链脲佐菌素(STZ)及高脂饮食的方法诱导2型糖尿病大鼠模型,应用离体小动脉灌流技术研究去除血管内皮后,累积浓度黄芪醇提取液对正常大鼠和糖尿病大鼠冠状小动脉平滑肌基础张力的影响;观察黄芪醇提取物对苯肾上腺素(PE)或KCl预收缩的微血管环的作用。结果 STZ和高脂饮食可致Wistar大鼠形成2型糖尿病,模型大鼠体质量与对照组比较明显减少,模型组大鼠与对照组大鼠相比血清胆固醇(TC)、甘油三酯(TG)和游离脂肪酸(FFA)差异具有统计学意义(P<0.05),糖化血红蛋白(HbAlc)和空腹血糖值明显增高,并出现高脂血症(P<0.05)。黄芪醇提取物对糖尿病大鼠冠状小动脉环有舒张作用;对PE和KCl预收缩的去内皮冠状小动脉产生浓度依赖性舒张作用。结论黄芪醇提取物对糖尿病大鼠冠状小动脉有舒张作用。
Objective To study the effect of alcohol extract of Astragalus on the tension of smooth muscle in coronary artery of diabetic rats. Methods The model of type 2 diabetic rats was induced by small dose of streptozotocin (STZ) and high-fat diet. The arterial perfusion of isolated arterioles was used to study the effects of different concentrations of extract of astragalus on normal rats And the basic tension of smooth muscle of coronary artery in diabetic rats. The effects of the extract of Astragalus on the pre-contracted microvascular rings of phenylephrine (PE) or KCl were observed. Results STZ and high-fat diet induced type 2 diabetes in Wistar rats. Compared with the control group, the body weight of model rats decreased significantly. Compared with the control group, the levels of serum cholesterol (TC), triglyceride ) And free fatty acid (FFA) were significantly different (P <0.05), HbAlc and fasting blood glucose were significantly increased, and hyperlipidemia (P <0.05). The extract of Astragalus membranaceus has a diastolic effect on the coronary artery rings of diabetic rats and a concentration-dependent relaxation of the PE and KCl precontracted endothelium-derived coronary arterioles. Conclusion Astragalus membranaceus alcohol extract can relieve the coronary arteries in diabetic rats.
